Match extender temperature to semen temperature and maintain them in a water bath; usually this is at 28 - 32°C ( Fig. 18 ). The antibiotics should also be maintained at the same temperature. The sample is observed under phase contrast microscopy at 200 - 400 x magnification and the percentage of sperm cells having progressive linear motion is determined. If the semen is too concentrated, the semen sample may be diluted with a buffered diluent or semen extender before coverslipping.
